چکیده

The Poly(A) binding (PAB) protein functions in posttranscriptional regulation by stabilizing mRNA and initiating translation. Using sequence homology to Arabidopsis PAB genes, we have identified seven members (OsPAB1 through OsPAB7 ) of the PAB gene family in rice. These can be divided into two groups based on sequence identity and expression pattern. The four genes in the first clade*/OsPAB2 , OsPAB4 , OsPAB6 , and OsPAB7 */share strong homology with AtPAB2 , and are expressed in all the organs that were examined here. The remaining three, OsPAB1 , OsPAB3 , and OsPAB5 , show the specific expression pattern for flowering. OsPAB1 is expressed throughout floral development, although its level decreases as the flowers mature. In contrast, expression of OsPAB3 is evident in older flowers and developing seeds, while OsPAB5 transcript is present only in mature flowers. All the genes in the second clade are expressed in carpels and stamens, but not in the paleae and lemmae of spikelets. In addition, we have identified a T-DNA insertional line, in which the gus reporter gene is fused to OsPAB1 to produce the PAB1-GUS fusion protein. In those experiments, GUS staining was strong in the ovaries and anthers, demonstrating that OsPAB1 is preferentially expressed in the reproductive organs. # 2003 Elsevier Science Ireland Ltd. Immunodetection of poly(A) binding protein II in the cell nucleus.

During the polyadenylation of pre-mRNA in vitro, poly(A) binding protein II (PAB II) binds to the growing poly(A) tail, stimulating its extension. The subcellular localization of PAB II was investigated with an antibody affinity-purified from rabbit serum raised against the purified protein. Immunofluorescence microscopy detected PAB II exclusively in the cell nucleus, both in a widespread stai...

Molecular Characterization of PAB 2 , a Member of the Multigene Family Coding for Poly ( A ) - Binding Proteins in A ra bidopsis f halia n a ’

l h e poly(A) tail of eukaryotic mRNAs associates with poly(A)binding (PAB) proteins whose role in mRNA translation and stabiiity is being intensively investigated. Very little is known about the structure and function of the PAB genes in plants. We have cloned multiple PAB-related sequences from Arabidopsis thaliana. Results suggest that PAB proteins are encoded by a multigene family. One memb...

Poly(A)-binding proteins are required for microRNA-mediated silencing and to promote target deadenylation in C. elegans

Cytoplasmic poly(A)-binding proteins (PABPs) link mRNA 3' termini to translation initiation factors, but they also play key roles in mRNA regulation and decay. Reports from mice, zebrafish and Drosophila further involved PABPs in microRNA (miRNA)-mediated silencing, but through seemingly distinct mechanisms. Here, we implicate the two Caenorhabditis elegans PABPs (PAB-1 and PAB-2) in miRNA-medi...

PAB-1, a Caenorhabditis elegans Poly(A)-Binding Protein, Regulates mRNA Metabolism in germline by Interacting with CGH-1 and CAR-1

Poly(A)-binding proteins are highly conserved among eukaryotes and regulate stability of mRNA and translation. Among C. elegans homologues, pab-1 mutants showed defects in germline mitotic proliferation. Unlike pab-1 mutants, pab-1 RNAi at every larval stage caused arrest of germline development at the following stage, indicating that pab-1 is required for the entire postembryonic germline deve...

Genomic organization, differential expression, and interaction of SQUAMOSA promoter-binding-like transcription factors and microRNA156 in rice.

Transcription factors play essential roles in the developmental processes of plants. Many such factors are regulated by microRNAs (miRNAs). SQUAMOSA (SQUA) promoter-binding-like (SPL) genes encode plant-specific transcription factors, some of which contain complementary sequences of miRNA156. In this study, 19 rice (Oryza sativa) SPL (OsSPL) genes and 12 rice miRNA156 (OsmiR156) precursors were...
